I'm doing an entirely new FSX install on a newly built computer. I'll be using the registered version of FSUIPC for FSX. In order to run Squawkbox on a second, networked, computer, do I run it through WideFS as I did with FS9? Thanks.

I think you use Squawkbox4 with FSX, and SB4 doesn't use FSUIPC4, only SimConnect (I think), so it won't use WideFS.
